How It Works:
Dexlansoprazole is a substituted benzimidazole antisecretory compound that suppresses gastric
acid secretion by selectively inhibiting the (H⁺, K⁺)-ATPase enzyme (gastric proton pump) in
parietal cells, blocking the final step of acid production.
Dosage:
Dexlansoprazole (as prescribed)
Side Effects:
Common: Diarrhea, abdominal pain, nausea, upper respiratory tract infection, vomiting,
flatulence
Less Common: Anemia, lymphadenopathy, angina, arrhythmia, bradycardia, chest pain,
edema, myocardial infarction, palpitation, tachycardia, ear pain, tinnitus, vertigo, goiter,
eye irritation/swelling, abdominal discomfort/tenderness, abnormal feces, anal
discomfort, Barrett’s esophagus, bezoar, abnormal bowel sounds, breath odor,
microscopic colitis, colonic polyp, constipation, dry mouth, duodenitis, dyspepsia,
dysphagia, enteritis, eructation, esophagitis, gastric polyp, gastritis, gastroenteritis,
gastrointestinal disorders
Drug Interactions:
Rilpivirine, atazanavir, nelfinavir, saquinavir, warfarin, methotrexate, digoxin, iron salts,
erlotinib, dasatinib, nilotinib, mycophenolate mofetil, ketoconazole/itraconazole, tacrolimus,
tetrahydrocannabinol (THC), CYP2C19 or CYP3A4 inducers (e.g., St. John’s Wort, rifampin),
CYP2C19 or CYP3A4 inhibitors (e.g., voriconazole)
Indications:
For patients aged 12 years and older:
Healing of all grades of erosive esophagitis (up to 8 weeks)
Maintenance of healing and relief of heartburn (up to 6 months)
Treatment of symptomatic non-erosive GERD heartburn (4 weeks)
When Not to Use:
Known hypersensitivity to dexlansoprazole or any excipient
Concurrent use with rilpivirine-containing products
Severe hepatic impairment
Precautions:
Use lowest effective dose for shortest duration to reduce risk
Increased risk of Clostridium difficile-associated diarrhea, especially in hospitalized
patients
Warnings:
1. Increased risk of osteoporosis-related fractures (hip, spine, wrist) with long-term PPI
therapy; manage at-risk patients accordingly
2. Monitor serum magnesium before long-term therapy, especially if using digoxin,
diuretics, or other hypomagnesemia-causing drugs
3. Prolonged treatment may cause vitamin B12 malabsorption and deficiency
