How It Works
Esomeprazole is a proton pump inhibitor (PPI). It binds irreversibly to the H⁺/K⁺ ATPase
enzyme system at the secretory surface of gastric parietal cells. This inhibits the final step of
acid production in the stomach, significantly reducing gastric acidity and promoting ulcer
healing and symptom relief.

Indications
 Treatment of GERD

When Not to Use
 Known hypersensitivity to esomeprazole or any other PPI

Precaution
Long-term use (especially >3 years) may result in vitamin B12 deficiency due to impaired
absorption.
Warnings
Warning 1:
Hypomagnesemia may occur with prolonged use and can lead to serious cardiac and
neuromuscular effects.
Warning 2:
Chronic PPI use may increase the risk of osteoporosis-related fractures (hip, spine, wrist).
Consider calcium and vitamin D supplementation.
Warning 3:
Avoid concurrent use with clopidogrel unless clinically necessary due to potential reduction in
clopidogrel’s effectiveness.
