β’ Β Β Valsartan: Blocks angiotensin II receptors β prevents vasoconstriction and aldosterone-mediated sodium retention β lowers blood pressure.
β’ Β Β Amlodipine: Calcium channel blocker (dihydropyridine) β inhibits calcium influx in vascular smooth muscle and myocardium β causes vasodilation.
β’ Β Β Hydrochlorothiazide: Thiazide diuretic β inhibits NaβΊ/Clβ» symporter in distal renal tubules β increases sodium and water excretion β reduces blood volume and blood pressure.

β’ Β Β Indicated for: Treatment of essential hypertension in adults, especially when monotherapy is inadequate.

β’ Β Β Hypersensitivity to valsartan, amlodipine, hydrochlorothiazide, sulphonamides, or dihydropyridines
β’ Β Β Pregnancy (2nd and 3rd trimester)
β’ Β Β Severe hepatic impairment, biliary cirrhosis, or cholestasis

o Β Β Renal artery stenosis (unilateral or bilateral)

1. Β Β Fetal Risk: Drugs affecting the renin-angiotensin system may impair fetal renal function β oligohydramnios, skeletal deformities, and neonatal death

3. Β Β Renal Impairment: Risk of acute renal failure, especially in susceptible patients

β’ Β Β Hematologic: Agranulocytosis, leukopenia, thrombocytopenia, hemolytic/aplastic anemia

β’ Β Β Neurological: Headache, hypertonia, visual disturbances
