1. Basic Information

2. Mechanism of Action
Pregabalin binds to the alpha-2-delta subunit of voltage-gated calcium channels in central
nervous system (CNS) tissues. This reduces calcium influx into neurons, potentially decreasing
the release of several neurotransmitters. This mechanism is believed to underlie its efficacy in
neuropathic pain, anxiety disorders, and certain types of seizures.
3. Indications
Pregabalin (Gabica) is indicated for:
 Neuropathic pain associated with diabetic peripheral neuropathy
 Postherpetic neuralgia
 Pain associated with spinal cord injury
 Adjunctive therapy for partial-onset seizures (with or without secondary generalization)
 Generalized Anxiety Disorder (GAD) in adults
 Fibromyalgia syndrome (FMS)
4. Dosage
 Active ingredient: Pregabalin
 Dosage to be determined by a healthcare provider based on indication and patient profile.
 Note: Taper off gradually over at least 1 week if discontinuing.
5. Contraindications
 Known hypersensitivity to pregabalin or any of the excipients
 Patients with hereditary galactose intolerance, Lapp lactase deficiency, or glucosegalactose malabsorption
6. Precautions
 Gradual withdrawal is recommended to minimize seizure risk.
 Use with caution in diabetic patients due to risk of weight gain.
 Monitor for signs of angioedema.
7. Warnings
1. Angioedema Risk: Discontinue immediately if facial, perioral, or upper airway swelling
occurs.
2. Weight Gain: Risk increases with dose and duration; may require adjustment in diabetic
treatment.
3. CNS Effects: May cause drowsiness, dizziness, or impaired coordination—caution
when driving or operating machinery, especially in elderly patients.
