⚙️ Mechanism of Action
Dexlansoprazole is a proton pump inhibitor (PPI). It suppresses gastric acid secretion by
inhibiting the H+/K+-ATPase enzyme at the gastric parietal cell surface, blocking the final step
in acid production.

🔄 Drug Interactions
 Contraindicated with: Rilpivirine, atazanavir, nelfinavir
 Caution with:
o Warfarin
o Methotrexate

o Erlotinib, dasatinib, nilotinib
o Mycophenolate mofetil
o Ketoconazole/itraconazole

📌 Indications
 Healing of all grades of erosive esophagitis (EE) – up to 8 weeks
 Maintenance of EE healing and heartburn relief – up to 6 months
 Symptomatic non-erosive GERD – 4 weeks treatment
(For patients aged 12 years and older)
❌ Contraindications (When Not to Use)
 Known hypersensitivity to dexlansoprazole or formulation excipients
 Co-administration with rilpivirine-containing products
 Patients with severe hepatic impairment
⚖️ Precautions
 PPIs may increase risk of Clostridium difficile-associated diarrhea in hospitalized
patients
 Use lowest effective dose for shortest necessary duration
🚨 Warnings
1. Fracture Risk: Increased risk of osteoporosis-related fractures (hip, spine, wrist) with
long-term/high-dose PPI use
2. Magnesium Deficiency: Consider serum magnesium testing before long-term use,
especially if on digoxin or diuretics
3. Vitamin B12 Deficiency: Long-term treatment may cause malabsorption of vitamin
B12
